Product Details:

  • Specificity: Detect both cellular and culture media (CM) high molecular weight (HMW) and low molecular weight (LMW) CHO proteins from various CHO strains such as CHO-S, CHO-K1, CHO-DG44, CHO-GS etc.
  • Immunogen: High molecular weight (HMW) and low molecular weight (LMW) CHO proteins
  • Purity: Antiserum or IgG. This antiserum was prepared by delipidation and defibrination followed by the addition of buffer salts and preservative.
  • Cross-Reactivity: Cross reactivity with other species is likely but has not been determined. Reactivity against homologues from other sources is not known.

Applications:

  • Applications: 1D/2D Western Blot, ELISA, IP
  • Application notes: Anti-CHO HCP antibody detects more than 2500 CHO total proteins with antibody coverage > 99% by 2D Western Blot. The antibody coverage in each quadrant is also > 99.4%.
  • Suggested dilutions: 1:1000 ~ 1:5000 for Western Blot, 1:2000 ~ 1:10000 for ELISA. Optimized dilutions may vary depending on testing samples and applications.

Handling

  • Precaution of use: This product contains Sodium azide: a POISONOUS AND HAZARDOUS SUBSTANCE, which should be handled by trained staff only.
  • Storage condition: Store at 4°C if using within 1-2 weeks. Upon delivery, aliquot and freeze at -20°C for extended storage. Avoid or minimize freeze / thaw cycles. Before using, bring to room temperature and centrifuge product if not completely clear.

A. CHO HCP antibody coverage by 2D Western Blot

150 µg HCP from CHO was labeled with Cy3 and run on large format 2D gel. After SDS-PAGE, the HCP image was scanned (top left), followed by transferring to membrane. The membrane was incubated with Anti-CHO HCP Antibody (CHO-S101) at a 1:2000 dilution at 4oC overnight. Cy5-labeled Donkey anti-Goat antibody was used for detection (top middle). Both CHO HCP (green) and WB (red) were scanned from the same membrane to generate the overlay image (top right).

B. CHO HCP antibody coverage by 1D Western Blot

20 µg HCP from CHO was labeled with Cy3 and run on a 1D gel. After SDS-PAGE, HCP image was scanned, followed by transferring to membrane. The membrane was incubated with Anti-CHO HCP antibody (CHO-G101) at 8 µg/mL final concentration at 4oC overnight. Cy5 labeled Donkey anti-Goat antibody was used for detection. Both CHO HCP (green) and WB (red) were scanned from the same membrane to generate the overlay image.

C. ELISA

The following data was generated using CHO-S101:

  • Coating: CHO-HCP protein in the indicated amount in 100 µl of PBS. 
  • Primary antibody: Anti-CHO HCP Antibody (CHO-S101) at 1:2000 dilution
  • Secondary antibody: HRP conjugated anti-goat IgG at 1: 5000 dilution
  • Substrate: TMB for 20 minutes.
